Method for the synthesis of pyrrole and imidazole carboxamides on a solid support
The present invention describes a novel method for the solid phase synthesis of polyamides containing imidazole and pyrrole carboxamides. The polyamides are prepared on a solid support from aromatic carboxylic acids and aromatic amines with high stepwise coupling yields (>99%), providing milligram quantities of highly pure polyamides. The present invention also describes the synthesis of analogs of the natural products Netropsin and Distamycin A, two antiviral antibiotics. The present invention also describes a novel method for the solid phase synthesis of imidazole and pyrrole carboxamide polyamide-oligonucleotide conjugates. This methodology will greatly increase both the complexity and quantity of minor-groove binding polyamides and minor-groove binding polyamide-oligonucleotide conjugates which can be synthesized and tested.

Pentapeptide with specific conformation, its production and use
Cyclic pentapeptides are disclosed having the following formula (I) --Cyclo(-A1 -A2 -A3 -A4 -A5 -)-- wherein A1, A2, A3, A4 and A5, are amino acid residues. The pentapeptide has amino acid residues in positions 1-2-3 to form a γ-turn, and amino acid residues in positions 3-4-5-1 to form a β-turn in combination with the γ-turn. D-α-amino acid residues are selected for A1, A3, and A5 and L-α-amino acid residues are selected for A2 and A4. Compounds having γ-turns and β-turns can be synthesized, regardless of the kinds of amino acid residues, and it is possible to synthesize compounds in which desired amino acid residues are introduced into sites of β-turn and γ-turn based on their importance from the viewpoint of biological activity. Wittig reagents and method for preparing α,β-unsaturated phosphonates
Wittig-type reagents and methods of preparation and use thereof for preparing α,β-unsaturated phosphonate esters from aldehydes and ketones are disclosed. The Wittig-type reagents have the following formulae: STR1 wherein X represents triflate, halide, BF4, SbF6, or ClO4 ; R1 represents alkyl, aryl or arylalkyl; and R2 represents alkyl, aryl or arylalkyl, provided that R1 and R2 not represent phenyl at the same time. The Wittig reagent diethyl phosphono-methylidenetriphenylphosphorane (1b) has been successfully synthesized for the first time via its phosphonium triflate salt (4a), by treating diethyl phosphonomethyltriflate with triphenylphosphine according to the disclosed method. The procedure has been applied to the synthesis of other new Wittig-type reagents such as phosphoranes and phosphonium salts. The new Wittig reagents thus synthesized were treated with various aldehydes and an activated ketone, affording the corresponding α,β-unsaturated phosphonates, saturated phosphonates or phosphoric acids. Triphenylphosphorane 1b and triphenylphosphonium 4a led to both cis and trans isomers with the latter being predominant, while trans isomers were almost exclusively formed when tributyl reagents (1c and 4d) were used.

PHOSPHONYL HYDROXYACYL AMINO ACID DERIVATIVES AS ANTIHYPERTENSIVES
Phosphonyl hydroxyacyl amino acids of the formula STR1 wherein X is a substituted or unsubstituted imino or amino acid or ester. These compounds possess angiotensin converting enzyme activity and are thus useful as hypotensive agents.
