- Cell in the mercury ion detection and for developing diphenyl ethylene bis-cyano benzene pair photon fluorescent probe

The invention relates to a diphenylethene group dual cyano benzene two-photon fluorescence probe for detecting and displaying intracellular Hg ions, and the probe is prepared by performing nucleophilic substitution on mercaptoethanol and 2,5-di(4-[di(2-chloroethyl)amino]styryl)para-phthalonitrile. Laser excitation of 810nm can avoid photoinduced toxicity of cells. In a range of pH being 4.5-13, the probe has good selectivity on the Hg ions, and Na, K, Ca, Mg and Mn ions have no interference with detection, and the Hg ions of micro molarity can be detected. Fluorescence intensity is reduced after complexing of probe molecules and the Hg ions, and the content of the Hg ions can be subjected to two-photon fluorescence detection. A two-photon fluorescence microimaging experiment shows that the probe has good permeability on fibroblasts and other cells, has no toxic or side effects on the cells and is particularly suitable for the detection of intracellular Hg ion concentration change and Hg ion distribution.

A novel two-photon fluorescence probe for Hg2+ derived from bis(styryl)terephthalonitrile, as a two-photon fluorophore, and bis[2-(2-hydroxyethyl sulfanyl) ethyl]amino group (ionophore), as a novel Hg2+ ligand, was developed. The probe possesses small molecule size, large two-photon absorption cross-section (1067 GM) in H2O, non-cytotoxic effect, long wavelength emission at 588nm, large Stokes shift (121nm), excellent photostability, high water solubility, good cell permeability, and pH insensitivity in the biologically relevant range. The probe can selectively detect Hg2+ ions in live cells and living tissues without interference from other metal ions and the membrane-bound probes, and its quenching constant is 8.73×105 M-1.

A new red-fluorescent mercury ion sensor material is designed and synthesized, which is composed of a tweezer-shaped hydrophilic probe containing bifurcated soft-base atoms N and S coupled with 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) unit. By virtue of the strong electron-accepting nature of TCF unit (as a push-pull chromophore), this designed sensor material can selectively detect Hg2+ over various tested metal ions in a 100% aqueous medium via naked-eye and photoluminescence (PL) observations. Theoretical and time-resolved photoluminescence measurements further confirmed the selectivity and reversibility of the probe towards Hg2+ via intramolocular charge transfer mechanism in this sensor material. Moreover, the living cell tests by confocal fluorescence images of this sensor material towards Hg2+ were also investigated. Finally, distinguished absorption changes and fluorescence quenching spectral appearances allowed us to present the selective optical indicator of Hg2+ via TCF moiety for the first time.

Two series of novel substituted 1,4-dihydropyridine derivatives incorporating nitrogen mustard pharmacophore hybrids without spacer DHP-M (4a-4d) and with ethyl spacer DHP-L-M (8a-8g) were designed and synthesized. They were subjected to in silico ADME prediction study to check their drug-like properties and evaluated for their cytotoxicity against: A 549 (lung), COLO 205 (colon), U 87 (glioblastoma), and IMR-32 (neuroblastoma) human cancer cell lines in vitro using 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay against chlorambucil and docetaxel. Majority of the test compounds exhibited moderate to significant cytotoxic activity. The highest activity in all the investigated cancer cells was displayed by DHP-M (4a). This may be due to the less steric hindrance offered by 4a.

Benzoic acid nitrogen mustard and its rare earth metal complexes were synthesized and characterized by elemental analyses, IR, electronic spectrum, and EPR. The interaction of synthesized complexes with Ct-DNA was investigated and reviewed as a mixed manner of both intercalation and alkylation via fluorescence titration. Their biological activities were also evaluated in K562 and Vero cell lines, indicating that complexes had a significant inhibitory effect; however, there was no synergistic effect instead of antagonistic effect compared to benzoic acid nitrogen mustard. The possible mechanism through cellular apoptosis was also explored by comet assay.

An organic chromophore (4-[N, N- bis(2′-chloroethyl)anilino] ethenyl N-methyl pyridinium-N- methyl iodide) L was synthesized and fully characterized by elemental analysis, IR, 1HNMR spectra and single crystal X-ray diffraction analysis. One and two-photon fluorescence spectra for it were studied. Interestingly, L exhibited obvious two-photon absorption in the range from 700-850 nm in DMSO solution. The interactions between the chromophore and calf thymus DNA were investigated by absorption, fluorescence spectra and viscosity experiments. The results showed that the chromophore could interact with DNA in the mode of intercalation and be effectively used as fluorescent staining dye for cell imaging applications. Furthermore, the cell viability data for MCF-7 (Human breast carcinoma) cells indicated that the low-micromolar concentrations of the L are essentially non-toxic and can be applied in brighter two-photon fluorescent (TPF) bioimaging.

A novel two-photon fluorescence probe for Zn2+ derived from dicyanostilbene as a two-photon fluorophore and 4-(pyridine-2-ylmethyl) piperazine as a novel Zn2+ ligand was developed. The probe shows a 72.5-fold fluorescence enhancement in response to Zn2+, a large two-photon action cross-section (580 GM), a noncytotoxic effect, and pH insensitivity in the biologically relevant range, and its dissociation constant (KdTP) is 0.52 ± 0.01 μM. The probe can selectively detect free Zn2+ ions in live cells for 1500 s or so and in living tissues at a depth of 80-150 μm without interference from other metal ions and the membrane-bound probes.

Due to the lipophilicity of the metal-ion receptor, previously reported Cu(I)-selective fluorescent probes form colloidal aggregates, as revealed by dynamic light scattering. To address this problem, we have developed a hydrophilic triarylpyrazoline-based fluorescent probe, CTAP-2, that dissolves directly in water and shows a rapid, reversible, and highly selective 65-fold fluorescence turn-on response to Cu(I) in aqueous solution. CTAP-2 proved to be sufficiently sensitive for direct in-gel detection of Cu(I) bound to the metallochaperone Atox1, demonstrating the potential for cation-selective fluorescent probes to serve as tools in metalloproteomics for identifying proteins with readily accessible metal-binding sites.
