- Monitoring intracellular pH fluctuation with an excited-state intramolecular proton transfer-based ratiometric fluorescent sensor

Intracellular pH is a key parameter related to various biological and pathological processes. In this study, a ratiometric pH fluorescent sensor ABTT was developed harnessing the amino-type excited-state intramolecular proton transfer (ESIPT) process. Relying on whether the ESIPT proceeds normally or not, ABTT exhibited the yellow fluorescence in acidic media, or cyan fluorescence in basic condition. According to the variation, ABTT behaved as a promising sensor which possessed fast and reversible response to pH change without interference from the biological substances, and exported a steady ratiometric signal (I478/I546). Moreover, due to the ESIPT effect, large Stokes shift and high quantum yield were also exhibited in ABTT. Furthermore, ABTT was applied for monitoring the pH changes in living cells and visualizing the pH fluctuations under oxidative stress successfully. These results elucidated great potential of ABTT in understanding pH-dependent physiological and pathological processes.

Herein, two kinds of structurally significant 2-(2-aminophenyl)benzothiazoles were selectively generated by the copper-catalyzed aerobic C?C bond cleavage of isatins. Morpholine served not only as a reactant but a promoter in the three-component assembly of 2-(2-aminophenyl)benzothiazoles. Also it proved to be an efficient additive for the formation of free amino 2-(2-aminophenyl)benzothiazoles. This protocol represents a step-economic entry to the title compounds with a range of compatible functionalities. (Figure presented.).

The invention belongs to the field of organic synthesis and metal catalysis, and discloses a method for preparing a 2-benzothiazole substituted aniline compound through catalysis of a copper compound.According to the invention, elemental sulfur which is cheap, easy to obtain, low in toxicity, easy to operate, stable and odorless is used as a sulfur source, commercially available isatin derivatives and 2-iodobenzidine are used as raw materials, and a series of 2-benzothiazole substituted aniline compounds are obtained at high yield through a cascade reaction under the catalytic action of a copper compound; 2-aminophenylmercaptan with unpleasant odor is prevented from being used as a raw material, and the invention has the advantages of simple reaction operation, mild conditions, easily available raw materials, short synthesis route and the like.

The invention discloses a ratio type fluorescent molecular probe as well as a preparation method and an application thereof, the structural formula of the ratio type fluorescent molecular probe is shown as a formula I. The preparation method of the ratio type fluorescent molecular probe for detecting the pH value is simple, raw materials are cheap and easy to obtain, the synthesis process is simple, the yield is high, and the ratio type fluorescent molecular probe is suitable for large-scale production, popularization and application. The fluorescent probe has excited state intramolecular proton transfer property, so that the interference of self-absorption and biological background fluorescence can be effectively avoided, and the error caused by non-uniform probe use concentration, non-uniform probe distribution and the like can be avoided due to ratio type response. In actual detection, the fluorescent probe is quick in response, wide in detection range and high in sensitivity, has high anti-interference performance on common coexisting substances in a sample, has good repeatability on detection of the pH value, and can realize repeated cyclic application by adjusting the pH value. The probe disclosed by the invention has a wide application prospect in the aspect of pH value detection in various scenes such as environmental samples, biological samples and the like.

The invention relates to polysubstituted benzothiazole and derivatives and a synthesis method thereof. According to the invention, catalysis with Cu is adopted for the first time, and in an oxygen atmosphere, an indole diketone compound, an o-aminothiophenol compound and morpholine are converted into N-(2-(benzo[d]thiazol-2-yl)phenyl)morpholine-4-formamide and derivatives, and the indole diketonecompound and the o-aminothiophenol compound are converted into 2-(benzo[d]thiazol-2-yl)aniline, so that the molecular structure is stable, and the chemical property is excellent. Reaction raw materials of the synthesis method are cheap and easy to obtain, pretreatment is not needed, and the atom economy of the reaction is high; only a cheap copper catalyst is used in the reaction, environmental pollution is reduced, raw materials are saved, and the reaction cost is reduced; the whole reaction system is simple, reaction conditions are mild, reaction equipment is few, experimental operation is simple and convenient, and material sources are wide.

The cyclocondensation of 2-aminothiophenol and carboxylic acids to give benzothiazoles has been carried out under mild conditions using tetrabutylammonium bromide (TBAB) as the reaction medium and triphenyl phosphite as the catalyst. Shorter reaction times, rapid isolation of the products, and excellent yields are advantages of this method. The reaction is found to be general and quite tolerant to the nature of the substituted carboxylic acids.
