The present invention provides compounds that can enhance functional O-mannosylation of proteins including alpha-dystroglycan. Also provided are methods of preparation of the compounds defined by the formula I. Also provided are the methods of using the compounds or the pharmaceutical acceptable salts or prodrugs thereof in treating and preventing subjects suffering from the diseases including muscular dystrophies and cancers.

- Synthesis method of 4,5,6,7-tetrahydrothieno[3,2-c]pyridine hydrochloride

The invention relates to a synthesis method of 4,5,6,7-tetrahydrothieno[3,2-c]pyridine hydrochloride. The method comprises the following steps: synthesizing 2-(2-thienyl)ethyl p-toluenesulfonate from 2-thiopheneethanol and paratoluensulfonyl chloride, further reacting the 2-(2-thienyl)ethyl p-toluenesulfonate with formamide to synthesize N-2-thienylethylformamide, carrying out cyclization on the N-2-thienylethylformamide under the action of trifluoroacetic acid to produce 6,7-dihydrothieno[3,2-c]pyridine, reducing the 6,7-dihydrothieno[3,2-c]pyridine with sodium borohydride, and carrying out salification to obtain the 4,5,6,7-tetrahydrothieno[3,2-c]pyridine hydrochloride. The invention provides a novel method for synthesizing the 4,5,6,7-tetrahydrothieno[3,2-c]pyridine hydrochloride. The method has the characteristics of low cost, low toxicity and simple technique, and has industrialized application prospects.

- Small molecules enhance functional O-mannosylation of Alpha-dystroglycan

Alpha-dystroglycan (α-DG), a highly glycosylated receptor for extracellular matrix proteins, plays a critical role in many biological processes. Hypoglycosylation of α-DG results in various types of muscular dystrophies and is also highly associated with progression of majority of cancers. Currently, there are no effective treatments for those devastating diseases. Enhancing functional O-mannosyl glycans (FOG) of α-DG on the cell surfaces is a potential approach to address this unmet challenge. Based on the hypothesis that the cells can up-regulate FOG of α-DG in response to certain chemical stimuli, we developed a cell-based high-throughput screening (HTS) platform for searching chemical enhancers of FOG of α-DG from a large chemical library with 364,168 compounds. Sequential validation of the hits from a primary screening campaign and chemical works led to identification of a cluster of compounds that positively modulate FOG of α-DG on various cell surfaces including patient-derived myoblasts. These compounds enhance FOG of α-DG by almost ten folds, which provide us powerful tools for O-mannosylation studies and potential starting points for the development of drug to treat dystroglycanopathy.

(Chemical Equation Presented) A series of Ugi reactions has been successfully performed using ammonia as the amine component, employing 2,2,2-trifluoroethanol as a non-nucleophilic solvent in order to suppress known side reactions. Utilizing concentrated aqueous ammonia as a convenient source, this approach offered a simple, one-step assembly of Ugi adducts suitable for elaboration into a variety of 5-aminoazole compounds through postcondensation modifications. Free or N-substituted 5-aminothiazoles and 5-(trifluoroacetamido) oxazoles were all prepared by this improved methodology. The scope of the synthetic route developed and application of the different products are discussed.

Novel peptide derivatives, compositions containing them, and their use for treating medical disorders resulting from a deficiency in growth hormone are disclosed. The peptides have the formula (I): STR1 wherein a, b, A, R1, L1, D, R3, R4, R2, L2, E and G are as defined in the specification. These peptides exhibit improved resistance to proteolytic degradation, and hence, improved bioavailability.
