Carbon-14 labeling of hydrochlorothiazide via [14C]formaldehyde production from [14C]carbon dioxide at ambient temperature
[14C]Formaldehyde was synthesized by reducing 14CO2 at ambient temperature with Schwartz's reagent. The [14C]formaldehyde was then used in the radiosynthesis of high specific activity (2.1 GBq/mmol) [14C]hydrochlorothiazide via cyclization of 4-amino-5-chloro-1,3-benzenedisulfonamide. A synthesis of [ 14C]formaldehyde by reducing 14CO2 at ambient temperature with Schwartz's reagent is described. Synthesis and stability of [1,3-14C2]-glyceryl trinitrate
The synthesis of [1,3-14C2]-glyceryl trinitrate ([14C]-GTN) labelled with quasi carrier free carbon-14 atom in the 1,3-positions of the molecule was effected in four reaction steps from 14CO2 ( 3,5% overall yield). The chemical and radiochemical purity of [14C]-GTN was found better than 99% controlled by HPLC-Radioactivity Monitor and TLC-Digital Autoradiograph methods. Study of the radiochemical stability in dilute solution at 5 °C shows l:hat the product can be stored at least for a month without detectable decomposition.
