Simplifying access to synthetic nucleosides is of interest due to their widespread use as biochemical or anticancer and antiviral agents. Herein, a direct stereoselective method to access an expansive range of both natural and synthetic nucleosides up to a gram scale, through direct glycosylation of nucleobases with 5-O-tritylribose and other C5-modified ribose derivatives, is discussed in detail. The reaction proceeds through nucleophilic epoxide ring opening of an in situ formed 1,2-anhydrosugar (termed “anhydrose”) under modified Mitsunobu reaction conditions. The scope of the reaction in the synthesis of diverse nucleosides and other 1-substituted riboside derivatives is described. In addition, a mechanistic insight into the formation of this key glycosyl donor intermediate is provided.

We describe a chemo-enzymatic synthesis of modified nucleosides through lipase-catalyzed hydrolysis of their peracetylated precursors. It was found from screening of a large number of substrates that these enzymesregioselectivities were affected by the sugar and the nucleobase structures. By selecting the best enzyme for each substrate in terms of activity and regioselectivity, we prepared a small library of differently monodeprotecled purine and pyrimidine nucleosides useful as intermediates for the synthesis of high-value nucleosides and mononucleotides. By this approach, the chemo-enzymatic preparation of doxifluridine (14) and uridine 5'-monophosphate (5'-UMP, 15) from peracetylated uridine 1 was carried out. Elimination of many of the processing stages associated with existing methods was achieved, and higher yields and products of increased purity were generated. Wiley-VCH Verlag GmbH & Co. KGaA.

- Stability and degradation kinetics of 5'-deoxy-5-fluoro-N4-(3,4,5-trimethoxybenzoyl)cytidine in aqueous solution

The stability of 5'-deoxy-5-fluoro-N4-(3,4,5-trimethoxybenzoyl)cytidine (Ro 09-1390) in buffered solutions at 37 °C was studied. Seven degradation products were identified by high-performance liquid chromatography (HPLC) and spectroscopy. The degradation products were different depending on the pH of the solution. Ro 09-1390 was unstable at pH 2.1 and stable at around pH 5.6 in spite of forming several decomposition products; and its degradation rate did not change in the pH range from 8 to 12. The degradation of Ro 09-1390 followed apparent first order kinetics and the decomposition pathway at alkaline conditions was different from the one at acidic conditions. On the other hand, at neutral pH the other decomposition products were formed.

The reaction of acetyl hypofluorite (AcOF) with unprotected uracil and cytosine nucleosides in acetic acid or water has been studied using (18)F as a tracer.For the nucleosides in general two cis-diastereoisomers of both the 6-acetoxy-5-fluoro and 5-fluoro-6-hydroxy adducts were obtained, (1)H n.m.r. analysis of which showed that they all possessed the anti-conformation.The 6-acetoxy-5-fluoroadducts in the uracil nucleosides showed a remarkable stability and appeared to be interesting versatile compounds.They could be converted into their hitherto unknown corresponding 5-fluoro-6-hydroxy-O6,5'-anhydrocyclouracil nucleosides.For the cytosine nucleosides the 6-acetoxy-5-fluoro adducts were not observed, while the other cytosine adducts were found to rapidly deaminate at C-4 in water yielding the corresponding uracil analogues.Interestingly, even within a pair of diastereoisomers different deamination rates were observed.
