Human 17β-hydroxysteroid dehydrogenase type 10 is a multifunctional protein involved in many enzymatic and structural processes within mitochondria. This enzyme was suggested to be involved in several neurological diseases, e.g., mental retardation, Parkinson’s disease, or Alzheimer’s disease, in which it was shown to interact with the amyloid-beta peptide. We prepared approximately 60 new compounds based on a benzothiazolyl scaffold and evaluated their inhibitory ability and mechanism of action. The most potent inhibitors contained 3-chloro and 4-hydroxy substitution on the phenyl ring moiety, a small substituent at position 6 on the benzothiazole moiety, and the two moieties were connected via a urea linker (4at, 4bb, and 4bg). These compounds exhibited IC50 values of 1–2 μM and showed an uncompetitive mechanism of action with respect to the substrate, acetoacetyl-CoA. These uncompetitive benzothiazolyl inhibitors of 17β-hydroxysteroid dehydrogenase type 10 are promising compounds for potential drugs for neurodegenerative diseases that warrant further research and development.

The method comprises the following steps of adding p-nitrophenol of nitrosonitric acid and sulfuric acid into phenol, adding methane chloride and anhydrous glacial acetic acid to obtain a mixture, then putting the mixture into a high-pressure autoclave, adding absolute ethyl alcohol and inputting hydrogen, performing pumping filtration to obtain filter residues after reaction, adding copper chloride into the filter residues, generating light green precipitation after adjusting pH through sodium hydroxide, filtering, washing the filter residues and drying to obtain the 2-amino-4-chloro-6-metoxyphenol.

- Nanocrystalline Pt-CeO2 as an efficient catalyst for a room temperature selective reduction of nitroarenes

We have developed a new synthesis strategy to prepare Pt nanoparticles with size between 2 and 5 nm supported on CeO2 nanoparticles with size between 30 and 60 nm by the hydrothermal method in the presence of the surfactant cetyltrimethyl ammonium bromide (CTAB) and a polymer (PVP). It was found that the catalyst is highly active for the chemoselective hydrogenation of nitro compounds in aqueous medium in the presence of molecular hydrogen at room temperature (25 °C). The catalyst was characterized by XRD, ICP-AES, XPS, BET-surface area measurements, SEM, TEM and EXAFS. Different reaction parameters like reaction time, catalyst ratio, Pt loading etc. were studied in detail. The investigation revealed that the site of Pt plays a crucial role in the activity by favouring the reduction of nitro-compounds. The catalyst shows >99.9% conversion of nitro-compounds with 99% selectivity of amino compounds. The reusability of the catalyst was tested by conducting the experiment with the same catalyst and it was found that the catalyst does not change its activity and selectivity even after five reuses. - Synthesis and anticoccidial activities of Quinoline Carboxylate derivatives with Methyl (E)-2-(3-methoxy)acrylate moiety

A series of novel quinoline carboxylate derivatives with methyl (E)-2-(3-methoxy) acrylate group were designed and synthesized as anticoccidial medicines. The structures were confirmed by 1H NMR, IR and HR-MS spectra. The biological activities were primarily evaluated according to the anticoccidial index method. The results indicated that these compounds (7c, 7d, 7e, 7g) exhibited anticoccidial activities against Eimeria tenella. In particular, the anticoccidial index of 6-decyloxy-7-ethoxy-4-{6-[2-(2-methoxy-1-methoxycarbonyl- vinyl)phenoxy]pyrimidin-4-yloxy}- quinoline-3-carboxylic acid ethyl ester (7e) was 168.7, which indicated that the compound has a good anticoccidial activity.

- Novel potent BRAF inhibitors: Toward 1 nM compounds through optimization of the central phenyl ring
-
BRAF, a serine/threonine specific protein kinase that is part of the MAPK pathway and acts as a downstream effector of RAS, is a potential therapeutic target in melanoma. We have developed a series of small-molecule BRAF inhibitors based on a 1H-imidazo[4,5-b]pyridine-2(3H)-one scaffold (ring A) as the hinge binding moiety and a number of substituted phenyl rings C that interact with the allosteric binding site. The introduction of various groups on the central phenyl ring B combined with appropriate A- and C-ring modifications afford very potent compounds that inhibit V600EBRAF kinase activity in vitro and oncogenic BRAF signaling in melanoma cells. Substitution on the central phenyl ring of a 3-fluoro, a naphthyl, or a 3-thiomethyl group improves activity to yield compounds with an IC50 of 1 nM for purified V 600EBRAF and nanomolar activity in cells.

N-(4-Hydroxyphenyl)-(5Z,8Z,11Z,14Z)-icosatetra-5,8,11,14-enamide (AM404) is a metabolite of the well-known analgesic paracetamol. AM404 inhibits endocannabinoid cellular uptake, binds weakly to CB1 and CB 2 cannabinoid receptors, and is formed by fatty acid amide hydrolase (FAAH) in vivo. We prepared three derivatives of this new (endo)cannabinoid using bioisosteric replacement (1), homology (2), and derivatization (3) of the 4-aminophenol moiety in AM404 and tested them against CB1, CB 2, and FAAH. We found affinities toward both cannabinoid receptors equal to or greater than that of AM404. Shortening the acyl chain from C 20 to C2 led to three new paracetamol analogues: N-(1H-indazol-5-yl)acetamide (5), N-(4-hydroxybenzyl)acetamide (6), and N-(4-hydroxy-3-methoxyphenyl)acetamide (7). Again, 5, 6, and 7 were tested against CB1, CB2, and FAAH without significant activity. However, 5 and 7 behaved like inhibitors of cyclooxygenases in whole blood assays. Finally, 5 (50 mg/kg) and 6 (275 mg/kg) displayed analgesic activities comparable to paracetamol (200 mg/kg) in the mouse formalin test.

The vanilloid receptor-1 (TRPV1 or VR1) is a member of the transient receptor potential (TRP) family of ion channels and plays a role in regulating the function of sensory nerves. A growing body of evidence demonstrates the therapeutic potential of TRPV1 modulators, particularly in the management of pain. As a result of our screening efforts, we identified (E)-3-(4-tert- butylphenyl)-N-(2,3-dihydrobenzo[b][1,4]dioxin-6-yl)acrylamide (1), an antagonist that blocks the capsaicin-induced and pH-induced uptake of 45Ca2+ in TRPV1-expressing Chinese hamster ovary cells with IC50 values of 17 ± 5 and 150 ± 80 nM, respectively. In this report, we describe the synthesis and structure-activity relationship of a series of N-aryl cinnamides, the most potent of which (49a and 49b) exhibit good oral bioavailability in rats (Foral = 39% and 17%, respectively).
