Pseudomonas aeruginosa (P.A.) is a human pathogen belonging to the top priorities for the discovery of new therapeutic solutions. Its propensity to generate biofilms strongly complicates the treatments required to cure P.A. infections. Herein, we describe the synthesis of a series of novel rotaxanes composed of a central galactosylated pillar[5]arene, a tetrafucosylated dendron, and a tetraguanidinium subunit. Besides the high affinity of the final glycorotaxanes for the two P.A. lectins LecA and LecB, potent inhibition levels of biofilm growth were evidenced, showing that their three subunits work synergistically. An antibiofilm assay using a double δlecAδlecB mutant compared to the wild type demonstrated that the antibiofilm activity of the best glycorotaxane is lectin-mediated. Such antibiofilm potency had rarely been reached in the literature. Importantly, none of the final rotaxanes was bactericidal, showing that their antibiofilm activity does not depend on bacteria killing, which is a rare feature for antibiofilm agents.

We report a rapid, sensitive and selective electrochemical sensor based on pillar[5]arene (CP5) reduced graphene (rGO) nanohybrid-modified glassy carbon electrode CP5-rGO/GCE for the trace detection of methyl parathion (MP) by differential pulse voltammetry (DPV) for the first time. Compared to beta-cyclodextrin (β-CD)-functionalized reduced graphene (rGO)-modified GCE β-CD-rGO/GCE, the proposed CP5-rGO/GCE sensor exhibits excellent electrochemical catalytic activity, rapid response, high sensitivity, good reproducibility and anti-interference ability towards MP. The recognition mechanism of β-CD/MP and CP5/MP was studied by 1H NMR. The results indicate a higher supramolecular recognition capability between CP5 and MP compared to that between β-CD and MP. The β-CD-rGO and CP5-rGO nano-composites were prepared via a wet chemistry approach. The resulting nano-composites have been characterized by thermogravimetric analysis (TGA), fourier transform infrared spectrometry (FTIR), charge transfer resistance (Rct) and zeta potential. The CP5-rGO/GCE combines the merits of CP5 and rGO, and is used for quantitative detection of MP. It has a low detection limit of 0.0003 μM (S/N = 3) and a linear response range of 0.001-150 μM for MP. This method has been used to detect MP in soil and waste water samples with satisfactory results. This study provides a promising electrochemical sensing platform and is a promising tool for the rapid, facile and sensitive analysis of MP.

The study of host-guest complexation between reactive 2-carboxyphthalanilic acid (CPA) and two cationic pillararenes has been carried out. Host-guest complexation with significant kinetic effects was observed only with the smaller cavity size pillararene (P5A). Kinetics in the pH range 1.50-6.40, ESI-MS, 1H NMR titration, and ROESY experiments were performed to characterize the complexes. High binding stoichiometry (H:G2) was observed for all CPA protonation states. The system is pH-dependent, and inversion of cooperativity (negative to positive) occurs by increasing the dianionic CPA2- concentration (allosteric behavior). Toward physiological pH, association constant K1:1 does not change (104 M-1), and K1:2 increased from 102 to 104 M-1, as well as the inhibitory effect increased up to 222-fold. NMR results elucidated the structure of the complex and allowed us to create a map of H-H interactions that describes well the diversity and number of interactions in the complex. ?

- Pillar[5]arene-based N-heterocyclic carbene ligand for Pd-catalysed Suzuki reaction

A pillar[5]arene-based N-heterocyclic carbene ligand was prepared by reaction of bromoethoxy pillar[5]arene with excess 1-methylimidazole at 130?°C in the absence of solvent and used as a catalyst for the Suzuki coupling reaction. Excellent yields were obtained when the Suzuki reactions were carried out under ambient atmosphere in ethanol, employing 0.2?mol% ligand, 1?mol% PdCl2(CH3CN)2 and 1.5?mmol of K2CO3. The novel pillar[5]arene-based imidazolium salt is a promising material for the construction of highly active supramolecular catalytic systems.

Pillar[5]arene-based 1-benzylimidazole and 1-(9-anthracenylmethyl)imidazole salts were prepared for the first time as novel class supramolecular NHC ligands. These ligands, together with PdCl2(CH3CN)2, were used as catalysts in the Heck coupling reactions of styrene and aryl halide derivatives. The 1-(9-anthracenylmethyl)imidazole salt exhibited better catalytic activity than the 1-benzylimidazole salt, owing to the steric bulk of the ligand. The novel pillar[5]arene-based imidazolium salt is a promising supramolecular skeleton for the construction of highly active catalysts.
