procedure of Towbin et al. (1979) and visualized by Pon-
ceau S dye. The 90 kDa protein band was cut out and 500
pmol protein was subjected to N-terminal sequence ana-
lysis by automated Edman degradation on a gas phase
sequencer (Model PSQ-2, Shimadzu, Japan). Phenylthio-
hydantoin (PTH) derivatives of individual amino acids
were identified by reverse phase HPLC.

The activity of pigeonpea urease was determined in
varying concentrations of substrate analogues like,
thiourea, hydroxy urea and methyl urea with hypo-
chlorite method (Weatherburn, 1967) where ammonia
released was monitored by reacting with phenol-hypo-
chlorite to form indophenol, due to their interference
with Nesslers’s reagent. Ki were determined by the
Dixon method. Km was calculated by Lineweaver–Burk
plot method.
