(isotopic distribution 35Cl, 75.78% and 37Cl, 24.22%). All samples were co-

immobilization of thiols and the tag was shown to be selective
for sulfhydryl-containing compounds. The resin was then
applied to a crude cell lysate of M. smegmatis to capture
mycothiol, demonstrating that metabolites can be detected
from complex samples at their native concentrations without
unmasking of this oxidation sensitive moiety. Importantly,
captured natural products can also be tracelessly released by
cleavage of the disulfide for direct isolation and
characterization. This MALDI-TOF MS-assisted strategy will be
broadly applicable for the discovery of compounds
immobilized on solid support and isolation of compounds
based on a variety of chemical functionalities by simply
switching out the terminal enrichment tag. Thus, it represents
a promising new method to explore natural product space.
