preparations—tablets (Sinequan, Pfizer, Germany) and cap-
sules, injections of doxepin hydrochloride (Doxepin, Pliva,
Poland). Table 4 shows the quantities obtained by the pro-
posed methods, official method, and labeled amount. The RSD
is lower than 1% indicating good precision and the indepen-
dence of the matrix effect over the absorbance measure-
ments. The results were compared with those obtained by offi-
cial BP method [5], which is based on UV-spectrophotometric
procedure, and found no to differ significantly.
The methods are advantageous when compared with the
reported methods which are tedious and involve expensive
experimental set up which ordinary laboratories cannot afford.
A significant advantage of the extractive spectrophotometric
methods are that they can be applied to the determination of
individual components in a multicomponent mixture. This
aspect of extractive spectrophotometric analysis is of major
interest in analytical pharmacy, since it offers distinct possi-
bilities in the assay of a particular component in complex
dosage formulations. In the present study, doxepin was esti-
mated successfully as a pure compound, as well as a compo-
nent in representatives of dosage formulations.

Doxepin (DOX) reacts with titanium (IV) and iron (III)
thiocyanate complexes forming a coloured ion-association
compounds were quantitatively extracted with a mixture of
butanol–chloroform (2:3) and (1:4), respectively. The prop-
erties were applied for the elaboration of new extractive spec-
trophotometric methods for determination of doxepin as a sub-
stance and in pharmaceutical preparations.
The methods enable the determination of DOX in the range
of 5–50 and 3–30 µg/ml in the examined DOX–Ti-SCN and
DOX–Fe-SCN systems, respectively. The proposed proce-
dures allow simple, fast and reliable quantitative analyses of
the drug which are always useful for routine determinations,
especially with the spectrophotometric procedures, which are
less time consuming, less expensive and less operational train-
ing. The proposed procedures can be used for the determina-
tion of doxepin in the presence of its degradation
product—dibenzo[b,e]oxepin-11(6H)-one. The precision and
reproducibility of the methods are good, RSD are low.
