5β,17α-Pregn-20-yno-1',2',5'-oxadiazol-17-ol (3) and its 5α-epimer (4) have been prepared for testing their anti-fertility profiles. 5β/α-Androstano-1',2',5'-oxadiazol-17-ones (5 and 6) needed for the purpose have been synthesised by two routes. 4,17β-Dihydroxy-4-androsten-3-one (10) obtained from testosterone is converted into its dioxime (11) which on heating with potassium hydroxide in ethylene glycol affords the oxadiazoles (12 and 13).Jones' oxidation of 12/13 furnish 5/6.In the other route, (25R)-3,4-dioximino-5ξ-spirostane (14) obtained from diosgenin is converted into spirostanoxadiazoles (15 and 16) which on Marker degradation, rearrangement of the 16-en-20-oximes and hydrolysis afford 5/6. 5/6 on ethynylation yield 3/4.
