Description
Valspodar, also known as PSC 833, is a non-immunosuppressant derivative of cyclosporine and a potent multidrug-resistance (MDR) modulator. It is a white solid that has the ability to restore the sensitivity of MDR cancer cells to cytostatic agents, making it a promising candidate for various pharmaceutical applications.

Valspodar is used as an ABCB1 (P-glycoprotein) inhibitor for overcoming multidrug resistance in cancer treatment. It inhibits the basal-to-apical transport and increases the apical-to-basal transport of certain cytostatic agents in cells that overexpress human P-glycoprotein (P-gp), thereby enhancing the effectiveness of these agents.
Valspodar is also used as an antineoplastic adjunct (chemosensitizer) to improve the efficacy of conventional chemotherapeutic drugs. It has been shown to increase survival time in MDR tumor-bearing mice and in an MDR leukemia mouse xenograft model when administered in combination with doxorubicin. Additionally, it prolongs the anti-hyperalgesic effects of intraperitoneally administered pregabalin in a mouse model of cold stress-induced central pain.
