The invention provides a preparation method of a 2-substituted-4-alkoxy imidazole compound, which comprises the following steps: (1) reacting 2-cyanoethylamine as shown in a compound 1 or a hydrochloride thereof with a compound 2, or reacting 2-cyanoethylamine hydrochloride with a compound 3 or a compound 4 to generate a compound 5 or a compound 6; and (2) reacting the compound 5 or the compound 6with sodium alcoholate shown as a formula I to obtain the 2-substituted-4-alkoxy imidazole compound shown as a compound 7. According to the invention, the 2-substituted-4-alkoxy imidazole compound issynthesized by using a simple and novel synthesis method, the route is short, the yield is high, the use of azide is avoided, the process is stable, and the industrialization is easy.
