The invention discloses a preparation method of N-2-pyridine-5-pyrimidine methylamine. The preparation method comprises the following steps: S1, reacting 2-aminopyridine with diethyl 2-bromomethylmalonate in an alkaline environment to obtain a substance 1; S2, hydrolyzing the substance 1 to obtain a substance 2; S3, enabling the substance 2 to react with N,N-dimethylformamide and phosphorus oxychloride, and then salifying a reaction product with sodium perchlorate to obtain a substance 3; and S4, enabling the substance 3 to react with formamidine and a condensing agent to obtain the N-2-pyridine-5-pyrimidinemethylamine. The invention provides a brand-new synthetic route, and the method has the advantages of accessible raw materials, low cost, simple and feasible operation, mild conditions,mature technique and higher yield and purity, and is suitable for industrial large-scale production.
