The invention discloses a one-pot method for preparing a substrate for detecting beta-D-glucuronidase. According to the method, specifically a compound with a structure of a formula (I) and acetobromo-alpha-D-glucuronic acid methyl ester are used as raw materials, the raw materials are subjected to a reaction for 8-24 h in the presence of a polar solvent and a coupling reagent, then an alkaline reagent is added, a reaction is carried out at room temperature for 4-8 h, then an acidic reagent is added to adjust pH to 2-3, and the substrate for detecting beta-D-glucuronidase is prepared through post-treatment. The method uses phenolphthalein with a substituted or un-substituted phenol structure, and acetobromo-alpha-D-glucuronic acid methyl ester as reactants to prepare the substrate for detecting beta-D-glucuronidase, reaction steps and operation are very simple and convenient, the process complexity is reduced, and thereby the expanded production is easy.
