Description

Tacrine Hydrochloride, also known as 1,2,3,4-tetrahydro-9-aminoacridine hydrochloride (THA, Cognex), is a reversible cholinesterase inhibitor that has been used in the treatment of Alzheimer's disease. It is a pale yellow solid with a bitter taste and is the first therapeutic launched specifically for Alzheimer's disease treatment. Tacrine functions as an inhibitor of both acetylcholinesterase (AChE) and butyrylcholinesterase, and it also inhibits the uptake of serotonin and norepinephrine in rat cerebral cortex. It is used to increase the levels of acetylcholine in patients with Alzheimer's disease, based on observations of reduced concentrations of ChAT and AChE in the brain.

Uses

Used in Pharmaceutical Industry:
Tacrine Hydrochloride is used as an anticholinesterase for the therapy of memory deficits in patients with Alzheimer's disease. It acts as a cognitive adjuvant by elevating the acetylcholine level in the cerebral cortex and slowing the degradation of acetylcholine from intact cholinergic neurons. Additionally, it selectively blocks potassium channels in the central nervous system, resulting in an increased release of acetylcholine and a prolongation of the action potential of the presynaptic cholinergic neurons.
Used in Alzheimer's Disease Treatment:
Tacrine Hydrochloride is used as a potent centrally acting anticholinesterase for the treatment of Alzheimer's disease. It has demonstrated clinically significant improvement in cognition in patients with the disease. Although the underlying cause of the dementing process is not altered, the beneficial effects of tacrine may be due to its multiple effects on several neurotransmitter systems.

Metabolism
