The method comprises the following steps of S1, preparing 1, 2-O-isopropylidene-3-O-acetyl-alpha-D-glucofuranose; S2, preparing 1, 2-O-isopropylidene-3-O-acetyl-5, 6-di-O-p-toluenesulfonyl-alpha-D-glucofuranose; S3, preparing 1, 2-O-isopropylidene-3-O-acetyl-5, 6-di-O-benzoyl-beta-L-idose; S4, preparing 1, 2-O-isopropylidene-L-idose, adding 100-200 mL of a newly prepared sodium methoxide-methanol solution of 0.1 M into a product obtained in the step S3, performing stirring at the normal temperature for 30-90 min, adding cation exchange resin to remove Na ions, performing filtering, and concentrating a filtrate under reduced pressure; and S5, preparing the L-idose.
