The invention relates to a preparing method of 2-(phenyl-substituted)-3-(benzoyl-substituted)indole, and particularly discloses a preparing method of 2-(3'-hydroxy-4'-methoxyphenyl)-3-(3'',4'',5''-triethoxybenzoyl)-6-methoxyindole (OXi8006). The structure formula of the OXi8006 is shown in the specification. The OXi8006 can generate good antitumor activity (IC50=1.1 muM) by inhibiting tubulin polymerization and is an important clinical candidate for treating tumor.
