Analgesic activity and metabolism in the mouse of morazone, famprofazone and related pyrazolones

The analgesic activity of morazone, famprofazone and some related pyrazolones as compared in mice. The synthesized pyrazolones were up to 5 times more potent than famprofazone. Methamphetamine or phenmetrazine were, with one exception, found in the urine of mice given the pyrazolones. The respective 4-formyl pyrazolones were found as additional urinary metabolites of the morazone analogues (C-4 phenyl ethylamine substituted), whereas the famprofazone analogues (C-3 phenylethylamine-substituted) gave 3-hydroxymethyl pyrazolones as urinary metabolites. Two other metabolites were isolated and identified as metabolites of two of the famprofazone analogues.
