Description

Cholesterol Esterase (CE) is a serine hydrolase enzyme that is released from the exocrine pancreas and belongs to the α/β hydrolase family of enzymes. It is a reversible enzyme that can hydrolyze or synthesize fatty acid esters of cholesterol and other sterols.

Uses

Used in Clinical Analysis:
Cholesterol Esterase is used as an enzymatic tool for the determination of total cholesterol when coupled with cholesterol oxidase.
Used in Simulated Digestion Models:
Cholesterol Esterase from porcine pancreas is used as a catalyst in an in vitro simulated digestion model to improve the hydrolysis of carotenoid esters in the intestinal phase and to analyze the bioavailability of phytosterol and cholesterol in the aqueous micellar phase.
Used in Enzyme Activity Assays:
Cholesterol Esterase from porcine pancreas is used as a standard in enzyme activity assays.
Used in γ-Oryzanol Hydrolyzing Activity:
Cholesterol Esterase from bovine pancreas is used for γ-oryzanol-hydrolyzing activity.
Used in Cholesterol Ester Quantification:
Cholesterol Esterase from bovine pancreas is used for cholesterol esterase enzyme activity in the quantification of cholesterol and cholesterol esters associated with tear film and lens samples.
Used in In Vitro Degradation Testing:
Cholesterol Esterase from bovine pancreas is used to test the in vitro degradation of poly(fumaroyl bioxirane) maleate (PFM) polymer designed for bone tissue engineering.

Biochem/physiol Actions

Cholesterol esterase (CE) is a reversible enzyme that can hydrolyze or synthesize fatty acid esters of cholesterol and other sterols. Hydrolysis of water insoluble long chain fatty acid esters requires bile salt activation. Hydrolysis of water soluble esters of short chain fatty acids and lysophospholipids does not require activation by bile salts. It also hydrolyzes tri-, di-, and mono-acylglycerols, phospholipids, lysophospholipids, and ceramides. The enzyme may have multiple functions in lipid and lipoprotein metabolism, as well as in atherosclerosis. Its molecular mass is approximately 129 kDa and the optimum pH range is 7.0-9.0. The enzyme is activated by cholic acid, glycocholic acid, BSA, Mg2+ and 0.3% (v/v) ? X-100. Ag+, Hg2+ and ionic detergents inhibit the activity of the enzyme.
