An efficient and simple source of nitroso reagents and their oxidation reactions are described. The combination of a Lewis acid and a metal nitrite was applied to the oxidation of silyl enol ethers. Amino acid and peptide derivatives were easily accessed through in situ C-C bond cleavage of fully substituted silyl enol ethers upon oxidation.

- Discovery and synthesis of tetrahydroindolone derived semicarbazones as selective Kv1.5 blockers
-
A novel class of tetrahydroindolone-derived semicarbazones has been discovered as potent Kv1.5 blockers. In in vitro studies, several compounds exhibited very good potency for blockade of Kv1.5. Compound 8i showed good selectivity for blockade of Kv1.5 vs hERG and L-type calcium channels. In an anesthetized pig model, compounds 8i and 10c increased atrial ERP about 28%, 18%, respectively, in the right atrium without affecting ventricular ERP.

Thionitrites or thionitrates have been considered to be unstable.However, bulky groups' thio-NOn such as t-butylthio-nitrites and -nitrates have been readily synthesized and found to be stable enough for the use of the useful organic syntheses as an excellent nitrosation and diazotization reagents under mild conditions.Direct conversion of amines to the corresponding halides in the presence of copper halides (II), fluorination of heterocyclic amines in the presence of sodium tetrafluoroborate, α- oximation of methylene groups in ketones, facile cleavage of C=N bond to ketones, and desulfurization of thioacetals and thioketals, and other useful organic syntheses are described.
