Dantrolene (CAS NO.7261-97-4) was first described in the scientific literature in 1967, proposed as a new class of muscle relaxant, and it was widely used in the management of spasticity and reported in a landmark 1975 article published in the British Journal of Anaesthesia.Its efficacy in humans was later confirmed in a large, multicenter study published in 1982.
Dantrolene (CAS NO.7261-97-4) is the only specific and effective treatment for malignant hyperthermia, a rare, life-threatening disorder triggered by general anesthesia, and is also used in the management of neuroleptic malignant syndrome, muscle spasticity, serotonin syndrome, ecstasy intoxication, and 2,4-dinitrophenol poisoning.

It also can be called Dantrium ; Hydantoin, 1-((5-(p-nitrophenyl)furfurylidene)amino)- ; and 2,4-Imidazolidinedione, 1-(((5-(4-nitrophenyl)-2-furanyl)methylene)amino)- . It gave positive results in animal high dose studies regarding mutagenicity and carcinogenity.And it depresses excitation-contraction coupling in skeletal muscle by binding to the ryanodine receptor, and decreasing intracellular calcium concentration. Dantrolene (CAS NO.7261-97-4) should not be given to breastfeeding mothers.
