298-83-9 Usage

Description

Nitrotetrazolium blue chloride (NBT) is a colorless to yellow dye that is reducible to blue or black formazan crystals by certain cells. It is a Tetrazolium dye used in colorimetric assays and can be used to assess cell viability as a function of redox potential. Intense blue dye used in histochemistry determinations, where actively respiring cells convert the water-soluble NBT to a deep blue-colored precipitate. It is a yellow crystal that is soluble in water, aqueous buffers, and dimethylformamide. Tetrazolium salts, like NBT, are useful in determining the metabolic activity of cells from various origins. The quaternary tetrazole ring core containing four nitrogen atoms, with a net positive charge, induces cellular uptake through plasma membrane potential, driving its use in cell biology.

Uses

1. Used in Cell Viability Assays:
Nitrotetrazolium blue chloride is used as a colorimetric indicator for assessing cell viability, as it changes color based on the redox potential within cells.
2. Used in Histochemistry Determinations:
NBT is used as an intense blue dye in histochemistry to detect actively respiring cells, which convert the water-soluble NBT to a deep blue-colored precipitate.
3. Used in the NBT Test:
Nitro Blue Tetrazolium is used in the "NBT test" to evaluate the activity of NADPH oxidase in phagocytes. This results in the production of blue reduced NBT formazan in normal cells but not in those from patients with chronic granulomatous disease.
4. Used as a Chromogenic Activity Stain:
NBT can be used as a chromogenic activity stain for oxidoreductases in gels or solutions.
5. Used in Western Blot Detection:
Nitrotetrazolium Blue chloride has been used as a substrate for western blot detection.
6. Used for Staining of Histological Tissue Sections:
NBT has been used for the staining of histological tissue sections.
7. Used in the Diagnosis of Chronic Granulomatous Disease:
Nitro Blue Tetrazolium chloride is used to confirm the diagnosis of chronic granulomatous disease.
8. Used as a Microscopic Stain:
NBT is a microscopic stain used to detect dehydrogenases and alkaline phosphatase.
9. Used as an NADPH-Diaphorase Substrate:
NBT acts as an NADPH-diaphorase substrate that competitively inhibits nitric oxide synthase.
10. Used in Immunology for Detection of Alkaline Phosphatase:
Nitrotetrazolium Blue chloride is used in immunology for the detection of alkaline phosphatase, often paired with 5-bromo-4-chloro-3-inolyl phosphate (PCIB) for the colorimetric detection of alkaline phosphatase activity. Alkaline phosphate converts PCIB to a product that reduces NBT to its formazan derivative, resulting in a black-purple precipitate.
Used in Pharmaceutical Industry:
Nitrotetrazolium blue chloride is used as a diagnostic tool for assessing cell viability and metabolic activity, which is crucial in the development and testing of new drugs and therapies.
Used in Biotechnology Industry:
NBT is used as a research tool in biotechnology for studying cellular processes, enzyme activity, and cellular respiration, contributing to the understanding of various biological mechanisms and the development of new biotechnological applications.

Hazard

Moderately toxic by ingestion.

Biochem/physiol Actions

Nitrotetrazolium blue (NBT), in combination with BCIP (5-bromo-4-chloro-3-indolyl-phosphate), is required for the detection of labeled probes in western blots. It is part of the colorimetric detection assay. NBT and BCIP dyes detect alkaline phosphatase activity, thereby generating an insoluble purple dye (diformazan). The dye is visible at areas where the labeled probe has hybridized to its target.

Safety Profile

Moderately toxic by ingestion. When heated to decomposition it emits toxic vapors of NOx and Clí.

Check Digit Verification of cas no

The CAS Registry Mumber 298-83-9 includes 6 digits separated into 3 groups by hyphens. The first part of the number, starting from the left, has 3 digits, 2,9 and 8 respectively; the second part has 2 digits, 8 and 3 respectively.
Calculate Digit Verification of CAS Registry Number 298-83:
(5*2)+(4*9)+(3*8)+(2*8)+(1*3)=89
89 % 10 = 9
So 298-83-9 is a valid CAS Registry Number.
